Abstract

Introduction. Modulation of cellular adhesion of the retinal pigment epithelium (RPE) is a highly promising therapeutic approach for different
eye diseases. As inflammation plays the crucial role in ocular diseases the impact of prostaglandins and proteases on adhesion of retinal pigment
epithelium seems to be possible.
The aim of the study. To evaluate the impact of prostaglandin F2α and urokinase plasminogen activator (UPA) on adhesion of primary retinal
pigment epithelium in culture.
Methods. Rabbit primary RPE cell culture was cultivated with prostaglandin F2α analogue (PA) 10-9 M and UPA 10-8 M. Cellular adhesion was
assessed after 24 h cultivation as well as cells number and epithelial–mesenchymal transition were assessed after 168 h cultivation.
Results. After 24 h cultivation PA and UPA groups revealed the 2.2 and 1.4 fold decrease in number of non adhesive cells, respectively, in
comparison with the controls. After 168 h cultivation PA and UPA groups revealed the 1.55 and 1.2 fold increase in cells number, respectively, if
compared with the controls. Moreover non control groups showed significantly higher epithelial mesenchymal transition.
Conclusion. PA and UPA in used concentrations have a prominent stimulating effect on the RPE cellular adhesion.
