Abstract

Introduction. Endometriosisis a commonovarianhormone-dependent diseasesin womenof reproductive age. Lack ofeffectiveness of treatmentof this diseasemay be associatedwith gapsof knowledgeabout the molecular mechanismsof its originand development. The aim of the study. In order to study the mechanisms of pathogenesis and molecular causes of differences between the clinical course of two histological types of ovarian endometrioid formations – glandular-cystic and cystic – we have performed clinical and laboratory researches and comparative analysis of gene expression of oestradiol and progesterone receptors in endometrioid ovarian tissue in 88 women of reproductive age. Methods. In biopsies unaltered ovary and endometrial tissue by PCR-RT we have determined the level of gene expression of oestradiol and progesterone receptors. Results. We revealed an increased expression of ERβ gene in both types of disease 3.3 times compared to normal tissues of the ovary (p<0,034), as well as increased expression of genes of two types of progesterone receptor: PR-A 18-fold (p=0,004) and PR-B in a 2,5fold (p=0,042) in the cystic ovary endometrial tissue type as compared with unmodified fabric. When comparing transcriptome of steroid receptors two histological types of ovarian endometriosis formations, we have found a difference in a single gene: expression PGRmC-1 4-fold higher in the cystic tissue type than in cystic glandular tissue type of ovarian endometriosis (p=0,038). Conclusion. We found aforementioned differences in the expression of steroid hormones, and it is not only clarify the mechanism of progesterone reception, but also clarify the possible molecular cause of more severe clinical course of glandular-cystic ovarian endometriosis type, compared with cystic type.
